The intersect of genetics, environment, and microbiota in asthma—perspectives and challenges
2021, Journal of Allergy and Clinical ImmunologyCitation Excerpt :Endotoxin has potent but paradoxical immunomodulatory effects and asthma associations,32,33 which may depend on penta-acylated versus hexa-acylated forms.34,35 The enzyme for hexa-acylated endotoxin, LpxM, is found exclusively in Gammaproteobacteria, which includes facultative pathogens Moraxella, Haemophilus, and Escherichia.34,36 Protection from asthma and allergy has been linked to overall microbial diversity in the living environment.17,37,38
Endotoxin predictors and associated respiratory outcomes differ with climate regions in the U.S.
2018, Environment InternationalCitation Excerpt :Several studies have reported that different endotoxin types may have different associations with asthma and wheeze depending on their chemical structure (Zhao et al., 2008; Norbäck et al., 2014). With regard to the number of acyl chains attached to the lipid A, some authors have speculated that penta-acylated endotoxin could be protective against allergic sensitization and IgE-mediated asthma, whereas the hexa-acylated form would increase the risk (Brix et al., 2014; Norbäck et al., 2014). The distinct effect of penta- and hexa-acylated endotoxin on inflammation and cytokine profiles has also been confirmed by animal studies in which a higher inflammatory response was associated with hexa-acylated endotoxin structure (Hađina et al., 2008).
