Materials and methods
The insertion of catheter and infusion of chondroitinase ABC after T8 complete spinal cord transection. We will use the SD female rats, 250–300 g weight, and all surgery was done under inhalation anesthesia with isoflurane and above homoeothermic blanket for keeping body temperature about 37 °C. Three different concentrations of chondroitinase ABC (ChABC, 1 U/ml, 5 U/ml) were applied to degradation of chondroitin sulfate proteoglycans (CSPG, a family of extracellular matrix, up-regulated following spinal cord injury) in complete spinal cord injury animal model. Discussion
Inhibition of axonal growth after SCI is thought to play a significant role in preventing regeneration and functional recovery. Some CSPG core proteins, particularly neurocan, versican, brevican, and NG2, are highly up-regulated following CNS injury [7]. In addition, other inhibitory factors such as myelin-associated inhibitors NoGo [8], and myelin-associated glycoprotein, could block axonal re-growth [8]. Intrathecal injection of ChABC after spinal cord injury upregulates regeneration-associated proteins, promote regeneration of sensory projections and corticospinal tract axons and behavior recovery (Bradbury et al., 2002; Huang et al., 2006). Mutant mice deficient in CS N-acetylgalactosaminyltransferease-1, necessary for CS synthesis, show complete functional recovery after spinal cord injury (Takeuchi et al., 2013).
