Original investigation: pathogenesis and treatment of kidney disease and hypertensionStem cell factor and crescentic glomerulonephritis

Patients
Twenty-eight of 43 available patients in the database of the Sheffield Kidney Institute (1992 to 2000), Sheffield, UK, were selected for the purpose of this study. Patients were excluded from the study either because of insufficient biopsy material or their biopsy specimens did not contain at least 10 glomeruli/biopsy. For a patient to have CGN, at least 30% of the total number of glomeruli in the biopsy should show crescents. Causes of CGN are listed in Table 1. Also included in the study are
Results
Median follow-up period for all patients was 8.25 months (range, 0.5 to 24 months). Demographic data for patients are listed in Table 2. Dividing patients into two groups based on the reciprocal of serum creatinine against time slope did not show a significant difference in clinical, histological, or immunohistochemical parameters of progressors and nonprogressors. However, in view of the short follow-up of some patients and that others presented with severely impaired kidney function that did
Discussion
This study shows for the first time the significant upregulation of the mast cell growth factor (SCF) in kidneys of patients with CGN. In this study, we confirmed previous observations of the upregulation of SCF in human nephropathies and showed the distribution to be primarily tubular, but also glomerular and interstitial. Also, we noted the presence of SCF within crescents. Previous studies have shown that mast cells, rather than SCF, may have a role in rapidly progressive GN1, 2 because they

Early prediction of renal graft function: Analysis of a multi-center, multi-level data set
2022, Current Research in Translational MedicineCitation Excerpt :Thus, there was a strong negative correlation of SCF serum levels with eGFR. An association between SCF and renal function has been observed repeatedly in other contexts [40–43]. Experiments in rat offer a potential cause, suggesting an association of SCF expression with renal fibrosis [44].
Mast cells in renal inflammation and fibrosis: Lessons learnt from animal studies
2015, Molecular ImmunologyCitation Excerpt :SCF serum levels rose importantly in patients with end-stage renal failure or patients with anti-neutrophil cytoplasmic autoantibody (ANCA) – associated GN (Kitoh et al., 1998) and, each time, levels were positively correlated with renal dysfunction parameters (El-Koraie et al., 2001). In addition, some authors found increased local concentrations of SCF in the kidney with a marked upregulation in patients with chronic renal failure (El-Koraie et al., 2001; El Kossi and El Nahas, 2003). SCF was found both in tubular cells and interstitial infiltrating cells as well as in the interstitial space and correlated with the number of mast cells.
Eosinophil activation of fibroblasts from chronic allergen-induced disease utilizes stem cell factor for phenotypic changes
2008, American Journal of PathologyCitation Excerpt :Evidence for a role of SCF in human airway disease was supported by studies demonstrating increased SCF expression in nasal polyps of asthmatics and allergic rhinitis patients.33,34 Additionally, SCF has been implicated as a potential mediator in fibrotic diseases in the kidney and intestine.34–37 Recently, the role of SCF has been verified using a chronic mouse model of allergic disease characterized by airway hyperresponsiveness, mucus overproduction, eosinophil accumulation, and peribronchial collagen deposition.11,13,14

2003, Kidney InternationalCitation Excerpt :The source and nature of MAPCs remains a subject of controversy and debate. Preliminary data from our laboratory and that of others [67] have also implied a contribution by bone-marrow-derived mesenchymal cells (MSCs) to the renal interstitial fibroblastic pool. MSCs are marrow-derived progenitors of tissue fibroblasts and shuttle through the circulation between the marrow and peripheral organs [39].
