Exploring the repertoire of IgE-binding self-antigens associated with atopic eczema
2009, Journal of Allergy and Clinical ImmunologyCitation Excerpt :After overnight growth, cDNA inserts of all picked clones were amplified by means of high-throughput PCR with a PTC-225 thermal cycler (Tetrad; MJ Research, Inc, Watertown, Mass) and gridded in duplicates onto filter membranes to produce hybridization filters. Twenty-four filter hybridizations with digoxigenin-labeled PCR probes derived either from known self-antigens or from the sequences of randomly selected clones were performed as previously described.25 Positive clones were scored with the image analysis package VisualGrid (GPC, Munich, Germany).
New computational approaches for analysis of cis-regulatory networks
2002, Developmental BiologyDifferential gene expression profiling of adult murine hematopoietic stem cells
2002, BloodCitation Excerpt :Peripheral blood cells were harvested at different time points after transplantation, as indicated in Figure 2C, and flow cytometric analyses were performed using different specific cell surface markers (CD3, B220, Mac-1, and Gr-1) together with anti-Ly5.2 antibody. In order to enrich for genes that are predominantly expressed in the uncommitted, or lineage-negative cell populations, and remove highly redundant and housekeeping genes for further analysis, high-density array18 (macroarray) analysis was used and cDNA clones that hybridized to the lineage-positive probes were removed. cDNA clones that had no or very weak hybridization signals to the lineage-positive probes were selected.
